Purification and Characterization of a Specific Gliadin Component from the Wheat Cultivar Marquis.

A specific gliadin component (band 50) was isolated from the wheat cultivar Marquis-K, purified, and characterized on the basis of its molecular weight, subunit composition, and isoelectric point. Purification was achieved using Sephacryl S-200 gel chromatography followed by preparative electrophoresis on a polyacrylamide gel. The preparation contains one gliadin, as shown by two-dimensional isoelectric focusing-polyacrylamide gel electrophoresis, is a monomer of molecular weight 53,250, and has an isoelectric point of 6.5.
